Description: Total Count provides an excellent nutrient base for general growth. This medium is used to determine total microbial count, without differentiation of microbial types. This medium may be used with regular or Tetrazolium-salt (T-salt) coated petri dishes. This medium used in conjunction with T-salt petri dishes will result in all bacterial colonies turning a red color, due to the reduction of the T-salt by the colonies. The red color makes counting easier for inexperienced users or when samples are very high in particulates. The function of the medium is unchanged when used with either type of petri dish. Total Count is also known as Aerobic Plate Count and Standard Plate Count. Must be used with Easygel® pretreated petri dishes. (included)

Composition: Tryptone, yeast extract, and glucose. pH 7.0 ± 0.2

Procedure: Standard Methodology for product being tested.

Incubation time and temperature:

24-48 hrs at 32-35°C.
48-72 hrs at ambient temperatures.

Dilutions should yield between 30-300 colonies per dish. This is for both accuracy and ease of counting.

Interpretation: On the regular petri dishes, non-pigment producers grow white to colorless. Those microbes which produce natural pigments grow in various shades. On the T-salt petri dishes, bacteria appear as pink to dark red units and are therefore easily spotted and counted.
